Chromium Linked to Telomere Damage

By Laura Hall
December 2009

On November 10, Patricia Opresko, Ph.D., an assistant professor at the University of Pittsburgh, presented "Links Between Telomere Instability, Environmental Genotoxins and Human Disease" for the Keystone Science Lecture Series at NIEHS. Opresko is an NIEHS 2006 Outstanding New Environmental Scientist (ONES) grantee. ONES is a program that funds early career scientists for five years to investigate the influence of the environment on human disease. Opresko’s research focuses on understanding telomere biology and the mechanisms of telomere loss.

During her talk, Opresko (http://www.biophysics.pitt.edu/site/faculty_detail.php?id=30)Exit NIEHS discussed telomere biology, the role of Werner syndrome helicase protein (WRN) and the deleterious effects of hexavalent chromium, Cr(VI), on telomeres. Human exposure to Cr(VI) occurs mainly as an occupational hazard in the welding industry where exposure is associated with respiratory problems and increased risk of lung disease. Cr(VI) is also found in pigments, anti-corrosive agents and sometimes at toxic waste sites.

Opresko explained that a telomere is a series of repetitive DNA elements at the end of a chromosome. Telomeres become shortened with each cell division, partly due to the inability of the DNA replicating enzyme to copy DNA at the tip of the telomere. Opresko said that this shortening mechanism normally limits the number of times cells can divide and this process is believed to be responsible for cellular aging and a limited lifespan. Sperm, egg and stem cells have an enzyme called telomerase that replaces the lost telomere sequence, while other cells of the body, called the somatic cells, do not show telomerase activity.

Fu-Jun Liu, Ph.D., a post-doctoral associate in Opresko’s lab, has looked at the effects of the environmental pollutant Cr(VI) and found that it causes replicative stress chromosome breaks due to replication or stalling fork breaks (see Environmental Factor article (http://www.niehs.nih.gov/news/newsletter/2009/october/science-protective.cfm)). WRN, which unwinds and separates double-strand DNA allowing the DNA to be copied or repaired, is involved in the recovery from this Cr(VI) damage.

These studies are "the first molecular evidence that WRN protects against replicative stress induced by environmental pollutants," said Opresko. "I think this is a real strength of the ONES program and the NIEHS strategic plan, to try to use environmental agents to inform about biological pathways."
In addition, Opresko and Liu found that Cr(VI) exposure causes telomere damage. Guanine residues of the telomeric repeats are particularly susceptible to oxidative stress caused by Cr(VI). The studies show that Cr(VI) induces chromatid breaks, telomere defects and telomere loss. Cr(VI) induces telomeric defects presumably by interfering with telomeric replication.

In Werner syndrome, a premature aging disease caused by mutations in the WRN gene, the number of sister chromatid exchanges increases relative to normal and is confined specifically to the telomeres. WRN appears to play a role in preventing exchanges, by regulating the recombination of the telomeres. Opresko is currently conducting studies to investigate if Cr(VI) exposure induces more telomeric defects in cells lacking WRN.

The research on Cr(VI) and telomere damage has led Opresko to examine the mutagenesis rate of replicating telomeres using an assay she generated in her lab. Opresko has found that under normal conditions this rate is very low. She plans to use this system to see which proteins are important in telomere replication.

Understanding the role and interactions of the genetic and environmental factors that affect telomere biology may help scientists understand aging and diseases such as Werner syndrome. Doing so may also help to identify groups or individuals that may particularly be at risk from certain occupational or other environmental exposures.

Expert Panel Recommends Listing Formaldehyde as Known Human Carcinogen

By Robin Mackar

Expert panel members Stephen M. Rappaport, Ph.D., and Lisa Peterson, Ph.D., listen intently as epidemiological studies are discussed. (Photo courtesy of Steve McCaw)

On November 4, a 10-member independent scientific expert panel convened by the National Toxicology Program (NTP) voted unanimously to list formaldehyde as a "known human carcinogen" in the upcoming 12th NTP Report on Carcinogens (RoC). Currently, formaldehyde is listed in the 11th RoC as "reasonably anticipated to be a human carcinogen." The vote came after three days of presentations, public comments and lengthy discussions of the body of literature on this widely used chemical.

"The decision to change the listing status of formaldehyde is based on sufficient evidence in human epidemiology studies, as well as in animal studies, and the fact that a substantial number of U.S. residents are frequently exposed to detectable concentrations of formaldehyde," said Kenneth E. McMartin, Ph.D., chair of the expert panel. McMartin is a member of the Department of Pharmacology, Toxicology and Neuroscience at Louisiana State University Health Sciences Center in Shreveport.

Formaldehyde is a high production chemical with a wide variety of uses. In the United States, formaldehyde is predominantly used to produce industrial resins that are used as adhesives and binders in wood products. It can be found in products such as particle board, glues, permanent press fabrics, paper product coatings, fiberboard and plywood. Formaldehyde is also commonly known for its use as a preservative in medical laboratories and mortuaries.
The panel reviewed numerous key epidemiological studies of workers exposed to formaldehyde in different occupational settings. The studies were deemed sufficient evidence to indicate a causal relationship between exposure to formaldehyde and cancer in humans. Taken together, these human studies revealed three types of cancers, including nasopharyngeal carcinoma, sinonasal adenocarcinoma and myeloid leukemia. "Chance, bias and confounding factors are unlikely to explain the observed excess in these cancers," McMartin noted.

The expert panel noted that in the case of myeloid leukemia, there is evidence of genetic damage in lymphocytes in the blood of formaldehyde-exposed people. These studies and toxicity studies in animals suggest that formaldehyde can produce effects at sites other than the nose and sinus that receive the initial contact of the colorless, strong-smelling gas.
"Having studies linking formaldehyde to not just relatively rare nasal cancers, but to a form of leukemia, which is a more common cancer, is an additional public health concern," said Ruth Lunn, Dr.P.H., (http://www.niehs.nih.gov/about/od/ontp/lunn/index.cfm) director of the Report on Carcinogens Center. (http://www.niehs.nih.gov/about/od/ontp/roc.cfm)
Formaldehyde is one of eight substances under consideration by the NTP for possible listing in the 12th RoC. Next, the NTP will solicit public comment on the expert panel's listing recommendation and scientific justification through the Federal Register, and finalize the background document, taking into consideration the panel's recommended edits and public comments. Following additional internal government review, the NTP will draft its listing recommendation for the 12th RoC and scientific information supporting the recommendation, solicit public comment on the listing recommendation, and convene a meeting of the NTP Board of Scientific Counselors to peer review the science supporting the listing recommendation.

Information about this meeting, and the review of formaldehyde or any other RoC nominated chemical, is available online. (http://ntp.niehs.nih.gov/index.cfm?objectid=03C9AF75-E1BF-FF40-DBA9EC0928DF8B15)

Dear Global Netters,

We are making a nice transition into the LinkedIn environment which has a new look and a new feel (see below a sample weekly digest).  As with any transition there are advantages and disadvantages.  One major advantage is that we will be able to network with a broader audience of environmental health and safety professionals.

Please notice that I had mirrored the regional sub-groups (Africa, Americas, Asia / Pacific Rim and Europe) under the GlobalOccHyg List on the LinkedIn Professional Network.  URL:  http://tinyurl.com/GlobalOccHyg-List

And to continue to provide a bridge from the Yahoo Groups, we set up RSS Feeds from each List.

I will see if I can come up with a conceptual diagram as we adjust and experiment with the information flow...

With the best holiday wishes,

Andrew Cutz

Evaluation of Endocrine Disruptor Screening Assays: Expert Panel Nominations and Data Submissions Requested

The National Toxicology Program Interagency Center for the Evaluation of Alternative Toxicological Methods (NICEATM) is requesting nominations of expert scientists for an international independent scientific review panel (Panel) to assess the validation status of two in vitro assays for their usefulness and limitations in determining whether chemicals interact with estrogen receptors in vitro. NICEATM is also requesting the submission of relevant data for consideration by the Interagency Coordinating Committee on the Validation of Alternative Methods (ICCVAM) and the Panel.

NICEATM will convene the Panel in collaboration with ICCVAM as part of the ICCVAM technical evaluation of the scientific validity of an in vitro stably-transfected estrogen receptor (ER) transcriptional activation (TA) Assay (LUMI-CELL® ER assay, XDS, Inc.) and an in vitro cell proliferation assay (CertiChem, Inc., MCF-7 Cell Proliferation assay).  All available data generated using these assays, or from in vivo or in vitro assessments for the 78 reference substances recommended by ICCVAM for the validation of in vitro ER and androgen receptor binding and TA test method will be considered.  Where appropriate, data will be used to assess the validity of the assays, and to aid in the development of performance standards for these test methods.

NICEATM invites the submission of relevant in vitro and in vivo data and information for the ICCVAM-recommended reference substances, or for other substances for which data exists from the LUMI-CELL ER assay and the CertiChem MCF-7 Cell Proliferation assay. NICEATM also requests nominations of scientists with relevant knowledge and experience to serve on the Panel.  Areas of relevant expertise include, but are not limited to, biostatistics, cellular biology, endocrinology, molecular genetics, regulatory toxicology, reproductive toxicology, and test method validation.

Information about the NICEATM-sponsored validation studies, as well as instructions on how to submit data and nominations, is available at:

ICCVAM has contributed to the approval or endorsement of 31 alternative safety-testing methods by Federal regulatory agencies since its establishment in 1997. Appropriate use of these test methods can significantly reduce animal use and improve animal welfare. ICCVAM has also identified critical research, development, and validation efforts needed to further advance numerous other alternative methods.

3rd INBR 2010 Congress, Abuja, Nigeria

7/24/2010 -> 7/30/2010

The 1st International Neuroscience Congress organized by the Institute of Neuroscience and Biomedical Research (INBR) was in July 2006, following the epoch-making SONA 2003 Conference, which held in Abuja.  INBR 2006 was very successful and attracted many participants from Nigeria, other parts of Africa, and outside Africa.  It was very remarkable that majority voted for a follow-up conference in 2008. It brought together people from different cultural, academic and research backgrounds. Collaboration was established and there was general awareness, great networking and mentoring. Many local scientists had the opportunity to visit the laboratories of foreign participants thereafter.

The 2008 INBR Congress was a great improvement over 2006 and it is hoped that 2010 will be a turning point for many scientists from within and outside Nigeria, in terms of great awareness of the real value of Africa in increasing the scientific, cultural and social content of the world. Increasing interest would be devoted to the young ones in making them think science in general and neuroscience in particular, as a career. The first champion of the International Brain Bee (IBB) Quiz contest will be concluded by then.

The INBR 2010 Congress will be a very important congress that will boost scientific research in Africa in particular and other parts of the world in general. So don't miss INBR 2010 Abuja, Nigeria.

Dear Colleagues,

For the last four years work has been taking place to develop an international system of training and qualifications for entry level occupational hygienists in developing countries. Consultation and discussions have been held at occupational hygiene conferences around the world and the system has been refined and developed to meet stakeholder needs.

The newsletter provides a brief update of recent developments, the organisations involved and the courses available.

Many thanks

Newsletter November 2009

Dear colleagues,

As previously, I am writing on behalf of the small group who have been progressing the development of this scheme. You are included in the distribution as you may have been involved in discussions, or expressed an interest at various workshops and presentations that have been made at occupational hygiene conferences worldwide over the last 3/4 years. We intend to provide periodic 'newsletters' but if you no longer wish to be included, please email ohta@... and your name will be removed from the list. Similarly, if you have received this indirectly and wish to be added to the list just email and let us know.

During the last few months there have been a number of significant and exciting developments. The name being used to describe the organisation handling the development of the international training modules is 'the Occupational Hygiene Training Association' (OHTA).  A company limited by guarantee has been registered under the name OHTA and rules of operation are being developed. This body will work under the guidance of IOHA and its member organisations and its board members will be drawn from IOHA and IOHA member organisations.

The success behind this project has been the cooperation between occupational hygiene bodies across the world. We share a common purpose and enthusiasm to promote the development of occupational hygiene programmes across the world through quality occupational hygiene training and qualifications at a range of levels. To support these aims there have been many workshops and discussions and agreements have been set out through a series of Memorandums of Understanding - a simple way to set out and demonstrate shared goals and ambitions to work together.

The original Memorandum of Understanding between IOHA members societies now has 10 signatures BOHS (UK), AIOH (Australia), AIHA (USA), OHSI (Ireland), SAIOH (South Africa), HKIOEH (Hong Kong), SOFHYT (France), MIHA (Malaysia), NYF (Norway) and ABHO (Brazil). <<Memorandum of Understanding .pdf>>

At the IOHA board meeting in October, the IOHA Board agreed to sign a Memorandum of Understanding with OHTA. As part of this IOHA consents to the use of the IOHA logo with the statement “Supported by IOHA” on certificates, website and training documentation. IOHA will also nominate a representative to the governing board

<<MOU-IOHA Signed 08 10 09 _2_.pdf>>

Together with AIOH and BOHS, the IOHA board have also agreed to provide funding toward the development of a website where the training materials can be hosted and accessed. Work is now progressing on the development of this learning portal to hold the materials and to provide links to participating IOHA member organisations and examining boards. The site will also have links to 'Approved' course providers. These are organisations who meet the basic criteria to run the courses and have been approved to offer the student assessment process. These links will enable site users to identify where and when courses are being offered. All course materials will be available free of charge and we hope to have the facility available by the year end. When the site is complete in March 2009, it will facilitate feedback and updating of hosted materials on a 'Wiki' style site.

One of the principles of this work is to improve the level of training in occupational hygiene and to provide a recognised scheme for qualifications at the intermediate/technician level - to complement the higher level professional qualifications recognised under the IOHA NAR scheme. NAR examination bodies are being consulted and, at the PCIH in Vancouver (October 2009), ABIH, the BOHS faculty of Occupational Hygiene and the  Canadian Registration Board of Occupational Hygienists signed an MoU to work together in the development of a common platform for credentialing individuals at this intermediate level. <<MOA.pdf>>

Work on the development and use of the scheme progresses well and we estimate around $500,000 has been invested so far in the development of training materials for 7 courses and the running of pilot courses around the world. The courses presently available are:

Module
W501 – Measurement of Hazardous Substances
W502
– Thermal Environment
W503
– Noise
W504
– Asbestos
W505
– Control
W506
– Ergonomics
W507
– Health Hazards of Substances

Each is a one-week taught course and the accompanying material includes a student manual, PowerPoint slides and tutor notes for all sessions, overnight student exercises, practical workshop sessions and student assessment.  A one week 'Fundamentals' course is also being developed to serve as a lower level introduction.

If you have an interest in participating in this project, running courses or working with others to set up courses, please let me know.

Finally, just to say, the work represents contributions, advice and support from many people in the global occupational hygiene community over the last 3 years – too many to mention here but thanks are due to all who have contributed.

Regards

AIHA’s Respiratory Protection Committee releases white paper on research needs in respiratory protection, By Sandy Smith, EHS Today, 11/24/09

Many current practices in respiratory protection are based on assumptions, past practices or extrapolation from laboratory studies, according to the American Industrial Hygiene Association (AIHA), and few studies have been done to evaluate the efficacy of or the need for each of these practices.

AIHA believes the results of research on the practical, applied topics presented in its newly released white paper may significantly enhance the safe and effective use of respiratory protection.

In recent years, the field of respiratory protection has experienced unprecedented, substantial opportunities and challenges. The establishment of the National Institute for Occupational Safety and Health (NIOSH) National Personal Protective Technology Laboratory (NPPTL) provides a focal point for research and standard setting for new classes of respiratory protective devices.  The increased demand for protection from chemical, biological, radiological and nuclear (CBRN) hazards; outbreaks of life threatening viruses; and new technologies for more precise characterization of aerosol particle sizes, respiratory hazards and respirator performance are generating increased interest in this subject area.

The AIHA Respiratory Protection Committee determined this is an opportune time to consider respiratory protection research topics and priorities. Having a very diverse membership, the committee established a project team consisting of users, government officials, manufacturers, researchers, standards developers and other interested industrial hygienists. The project group identified seven high priority research topics including:

(1) Measurement of respirator performance
(2) Qualitative fit testing for full facepiece respirators
(3) Appropriateness of fit factor safety margin criterion
(4) In-facepiece measurements
(5) Effectiveness of respiratory protection program requirements
(6) Effectiveness of user seal checks
(7) Organic vapor cartridge desorption

AIHA hopes that NIOSH, universities and independent researchers will use this list to further investigate the benefits and to improve available respiratory protection.

What You Need to Know About the Safe Use of Spray Polyurethane Foam

Spray Polyurethane Foam (SPF) is a highly effective weatherization
product that is playing an important role in our nationwide effort to
dramatically increase the energy efficiency of our homes, schools and
buildings. Spray applicators and other on-site workers, as well as
Do-it-Yourself applicators and the consumer, need to understand that
proper protective equipment and other precautions are essential to safe
installation of these materials, and that without proper protections,
exposures to the spray during installation could result in asthma and
other health effects.

The Environmental Protection Agency is pleased to host a special federal
and industry panel discussion on the safe use of SPF.

An expert panel will address cutting-edge best practices and emerging
SPF issues of interest to the building, construction, and weatherization
industries; government officials, including school administrators; and
consumers.

A question and answer session with the federal and industry experts will
follow the panel presentations.

Panel participants include:
- U.S. Environmental Protection Agency
- U.S. Occupational Safety and Health Administration
- National Institute for Occupational Safety and Health
- U.S. Consumer Product Safety Commission
- American Chemistry Council’s Center for Polyurethanes Industry
- Spray Polyurethane Foam Alliance

EPA will be hosting a Webinar on Wednesday, December 2, 2009 (1-3 PM
EST). For more information and to register for the Webinar, please visit

ACGIH is pleased to present the following webinar:

Ethical EH&S: Effective Implementation of Corporate Social Responsibility in the
Global Economy

Date: LIVE on December 10, 2009
Time: 2:00 pm, Eastern Standard Time (GMT -05:00, New York)
Registration and Course Information Link:

Course Content:

This webinar will fulfill both the applicant and Diplomat requirement for at
least 2 hours of ethics education required by the American Board of Industrial
Hygiene (ABIH). One the biggest ethical challenges for occupational and
environmental health professionals today is ensuring that corporate supply
chains in the global economy effectively protect the health of workers,
surrounding communities, and the environment. Numerous reports by the media and
non-governmental organizations (NGOs) over the last 15 years have documented how
even first-tier suppliers for international "brand name" companies have
frequently operated unsafe, unhealthy, and polluting factories. The conditions
in second or third tier suppliers' plants are worse. The current global economic
crisis threatens to make matters worse as cost-cutting measures can accelerate a
"race to the bottom" in factory conditions and the adjacent communities. Webinar
speakers will present a comprehensive perspective on meeting industrial
hygienists' ethical responsibilities in an effective, sustainable and profitable
manner. Presentations will focus on:

~ how a "triple bottom line" approach fulfills companies' ethical
responsibilities and adds measurable value to business operations.
~ how effective workplace safety in global supply chains is only possible with
the active involvement of informed and empowered workers playing a direct
role in plant-level OH&S programs.
~ the biggest failures of corporate social responsibility programs over the last
decade, and on what human rights NGOs expect from occupational health
professionals and the companies that employ them.

Presented by - Glenn Barbi, MS, CIH; Garrett Brown, MPH, CIH; and Lynda Yanz

To attend this webinar, you must first register for it. Once you have
registered, you will receive an email message confirming your enrollment status

Andrew

As you know, many organizations have been facing a number of critical transformation initiatives, including:

-         Organizational streamlining

-         Total organizational transformation

-         Acquisition merger integration

-         Financial reporting and systems implementation / transformation

-         IT implementation

-         New service, program or product launch

-         Strategy implementation

Transformation programs such as these consume vast amounts of internal resources, are complex from both a technical and organizational culture perspective, and involve a high degree of risk. Unfortunately however, many of these transformation initiatives fail to deliver on expected outcomes. What is often lacking is both the art and science of transformation management.

Within this context, attached is a light easy read on “Assessing the Art and Science of Project Management” just published in the November edition of PM World Today.

Hope you enjoy it.

I would also invite you to take a look at the attached pdf brochure [left click to advance to the next page, right click to reverse, ESC to end]

If you are interested, I’d be happy to chat further by telephone; just let me know what works best.

Wishing you a great day!

Cheers
