Effect aspartame

THE EFFECT OF ASPARTAME TO THE SERUM CREATININE LEVEL AND HISTOLOGIC STRUCTURE
OF KIDNEY IN MICE (Mus musculus L.) STRAIN SWISS

ABSTRACT

The aims of this research were to know the effect of aspartame to the serum
creatinine level and histologic structure of kidney (glomerules and proximal
tubules).

This research was evaluated experimentally to male mice with average weight of
20 gr.

These mice were divided randomly in to 5 groups i.e.: control, 5 mg aspartame,
10 mg aspartame, 15 mg aspartame, and 20 mg aspartame.

The treatment was done for 28 days, before treatment blood had taken and
measured creatinine serum level.

After treatment mice blood had taken again, killed and sectioned kidney organ to
make histologic preparation.

Creatinine serum level, wide average Bowman space glomeruli and lumen proximal
tubules was analyzed
using analysis of varians (ANOVA) and continued Duncan Multiple Range Test
(DMRT) on 5% significans,
time factor of creatinine serum level was analyzed using T-test.

Observation of kidney histologic structure (glomerules and proximal tubules)
were done to determine the kind and level of damage and analyzed descriptively.

Creatinine serum level control normaly,
dosage variation aspartame creatinine serum level increased up to ± 3.29 mg/dl.

There were control glomerules and proximal tubules normaly.

Dosage variation aspartame glomerules damage, i.e.:
cloudy swelling degeneration; karyorectic; picnotic; karyolitic; wide Bowman
space and hemolitic erytrocyte,
proximal tubules damages were picnotic; karyorectic; karyolitic; hydrophic
degeneration; lumen narrowed and protein cast.

It appears that you find toxic effects to the kidney at the lowest dose of
250 mg aspartame per kg body weight.
Mice and rats are about ten times more resistant to methanol (formaldehyde)
toxicity than humans.

How many mice did you test?

You may choose to use Dimethyl Dicarbonate as a palatable source of methanol
to do the same study to find if methanol is the part of aspartame that damages
kidney tissues in mice:

One liter aspartame diet soda, about 3 12-oz cans,
gives 61.5 mg methanol,
so if 30% is turned into formaldehyde, the formaldehyde
dose of 18.5 mg is 37 times the recent EPA limit of
0.5 mg per liter daily drinking water for a 10-kg child:

Dark wines and liquors, as well as aspartame, provide
similar levels of methanol, above 120 mg daily, for
long-term heavy users, 2 L daily, about 6 cans.

Within hours, methanol is inevitably largely turned into
formaldehyde, and thence largely into formic acid -- the
major causes of the dreaded symptoms of "next morning"
hangover.

Fully 11% of aspartame is methanol -- 1,120 mg aspartame
in 2 L diet soda, almost six 12-oz cans, gives 123 mg
methanol (wood alcohol). If 30% of the methanol is turned
into formaldehyde, the amount of formaldehyde, 37 mg,
is 18.5 times the USA EPA limit for daily formaldehyde in
drinking water, 2.0 mg in 2 L average daily drinking water.
