Hello everyone, I just got my notice to make my appointment with my
hemtologist Dr. in March.
I can't beleive it will my 3 year aniversary with C.M.L.
I'am a single 400(gleevec) pill per day since diag. 3/02
I've been in remission since my 6 month(9/02) check up by F.I.S.H.
Test. I have my CBC'S checked 1 x month.
My last two 6 month check ups(3/04)& (9/04) have been by RT-PCR TEST.
I'am currently in Molecular Remission by RT-PCR TEST.
Age 46 w/m law enforcment.
Again, for those you are considering a bone marrow
transplant..consider gleevec first.Mike

My onc says, if he had to consider bone marrow transplant for me, that would be
his and my last resort. He says since Gleevec works so good for me, he will not
give up on me.
I am doing well so far. One never knows how the body will accept the new imune
system, I would rather not do it. Sounds very harsh, but thats how I feel.

Hi Kim,

I've been on Gleevec, 400mg, for almost 4 years, and I also get
cramps and sometimes feel 'achy' all over.  It helps me to drink a
lot of water, eat a banana and/or orange for breakfast in the morning
(for potassium) and I take a calcium/magnesium pill in the afternoon.
It's best to take the calcium/magnesium combination, because calcium
needs magnesium to work and absorb properly.

Some moderate exericise helps too, like a 20 minute daily walk.

Linden.
I also have foot cramps which at times are nearly unbearable.No one seems to
know what causes them.My endocrinologist prescribed potassiun for me but the
drugist said I shouldn't take them with gleevec.I used magnesium for a while til
that stopped working.I have been at my wits end because I don't seem to be able
to get rid of the cramps which have recently started in my legs and hips.
Whether it's gleevec related I don't know.

Kim,
My mom has been on gleevec for just about a year for GIST (a rare form of gastro
cancer). She is on 800 mg and suffered from severe leg cramps. With the help of
a potassium pill and orange the cramps have gotten a lot better. Magnesium and
Calcium was also recommended to her to help with the leg cramps, but potassium
seems to do the job for her.

FDA Clears First of Kind Genetic Lab Test
The Food and Drug Administration (FDA) today cleared for marketing
the first laboratory test system that will allow physicians to
consider unique genetic information from patients in selecting
medications and doses of medications for a wide variety of common
conditions such as cardiac disease, psychiatric disease, and cancer.

"Physicians can use the genetic information from this test to prevent
harmful drug interactions and to assure drugs are used optimally,
which in some cases will enable patients to avoid less effective or
potentially harmful treatment choices," said Dr. Lester M. Crawford,
Acting FDA Commissioner.

The new test is the AmpliChip Cytochrome P450 Genotyping Test made by
Roche Molecular Systems, Inc., of Pleasanton, Calif. It was cleared
for use with the Affymetrix GeneChip Microarray Instrumentation
System, manufactured by Affymetrix, Inc., of Santa Clara, Calif.

The new test is the first DNA microarray test to be cleared by the
FDA and its clearance paves the way for similar microarray-based
diagnostic tests to be developed in the future. A microarray is
similar to a computer microchip, but instead of tiny circuits, the
chip contains millions of tiny DNA molecules. The test is performed
using DNA that is extracted from a patient's blood. A person's DNA
sequence is determined based on the sequence of the probe molecule to
which the DNA is most similar.

The new test analyzes one of the genes from a family of genes called
cytochrome P450 genes, which are active in the liver to break down
certain drugs and other compounds. Variations in this gene can cause
a patient to metabolize certain drugs more quickly or more slowly
than average, or, in some cases, not at all. The specific enzyme from
this family that is analyzed by this test, called cytochrome P4502D6,
plays an important role in the body's ability to metabolize some
commonly prescribed drugs including antidepressants, anti-psychotics,
beta-blockers, and some chemotherapy drugs.

The test is not intended to be a stand-alone tool to determine
optimum drug dosage, but should be used along with clinical
evaluation and other tools to determine the best treatment options
for patients.

FDA cleared the test and the scanner based on results of a study
conducted by the manufacturers of hundreds of DNA samples as well as
on a broad range of supporting peer-reviewed literature.

AG-858 personalized cancer vaccine
An investigational product in clinical trials
What is AG-858?
AG-858 (HSPPC-70) is an investigational personalized vaccine designed
to treat cancer while minimizing side effects that are often
associated with other treatment options. AG-858 is currently being
investigated as a cancer treatment and is only administered to
patients in research studies.

How does AG-858 work?

AG-858 is a vaccine made from individual patients' cancerous cells.
Patients undergo a blood filtering process called leukapheresis,
during which white blood cells are collected. The white blood cells
are sent to Antigenics so that the personalized vaccines can be made.

Vaccines provoke the body's natural disease-fighting response. A
vaccine that is made from the patient's own cancerous cells is
designed to contain the particular cancer's `fingerprint.' Injection
of the AG-858 vaccine may cause the body to attack any cells bearing
this cancer fingerprint.

How is AG-858 different from Oncophage and other treatments?
AG-858 is based on a different heat shock protein than the one used
in Oncophage. AG-858 is a vaccine based on heat shock protein 70
(HSP70), whereas Oncophage is based on a heat shock protein called
gp96.

What indications for AG-858 are being tested?
AG-858 is currently being evaluated in combination treatment for
chronic myelogenous leukemia (CML). Based on findings from an ongoing

earlier HSP70-based vaccine in combination with the FDA-approved
therapy Gleevec® (imatinib mesylate, Novartis), Antigenics is
conducting an exploratory Phase 2 study of AG-858 in combination with
Gleevec for the treatment of chronic phase CML.

How is AG-858 administered?
In the trial of AG-858 in chronic myelogenous leukemia, white blood

Hi, I am writing on behalf of my mother who has been on Gleevec for a few
years. She has always had on again/off again leg cramps on the drug, but
recently they have become significantly worse.

She says that after she sits or lays down, when she tries to stand up every
bone in her body hurts. She's a very active 58 year old who suddenly feels
like a 90 year old.

We are trying to figure out whether this might be Gleevec related. Has
anyone else had comparable experiences?

- Next-generation leukemia pills, designed to help patients not cured
by the successful drug Gleevec®, work even better than doctors had
hoped, researchers said on Sunday.

One new drug, made by Bristol-Myers Squibb, put 86 percent of
patients who tried it into remission - meaning signs of their cancer
disappeared, the researchers said.

Although they were only Phase I trials, meant to show the drug was
safe, the effects were dramatic, the doctors told a meeting in San
Diego of the American Society of Hematology.

"Certainly it is wonderful. It will save lives," said Alan
Kinniburgh, Ph.D., senior vice president, Research Administration,
The Leukemia & Lymphoma Society.

Oncologists hope the approach may work in many other cancers, too.
Dr. Kinniburgh said it is likely that the new pills will be used in
combination, like old-fashioned chemotherapy.

"I know of no cancer where one single drug has ever cured the
cancer," he said.

The new drugs are being tested in patients with chronic myeloid
leukemia (CML), which affects about 4,400 Americans a year and 10,000
people around the world.

High Remission Rate
The Bristol drug is known by its experimental name BMS-354825. During
the trial, also financed by Bristol-Myers, 31 of 36 patients with
advanced CML, who had not been helped by Gleevec, had a complete
hematologic response, meaning their bodies stopped producing leukemia
cells.

This translates to an 86 percent remission rate, said Dr. Charles
Sawyers, a Howard Hughes Medical Institute investigator at the
University of California Los Angeles, who is helping test the drug.

Gleevec, made by Swiss drug company Novartis and marketed as Glivec
in Europe, targets an enzyme, called BCR-ABL, that leukemia cells use
to proliferate. The drug attaches to the cancerous cells and stops
them from growing and spreading. It was the first "targeted" cancer
drug and made headlines when it was approved in 2001 because never
before had a simple pill shown such dramatic effects in cancer.

Gleevec, or imatinib, is Novartis's second-biggest product, with
sales in the first nine months of this year of $1.1 billion. But in a
few patients, perhaps 12 percent, the cancer cells mutate just enough
to slip out of Gleevec's grip. The cancer comes back.

New Pill Seems Safe
So some of the researchers who worked on Gleevec teamed with Bristol-
Myers Squibb to develop the new pill, which is less picky about how
it grabs onto a cancer cell to deactivate it. The Bristol-Myers drug
affects a different enzyme called SRC, pronounced "sark."

Sawyers said he and colleagues worried that there could be unforeseen
side effects in patients, as no one had ever tested a SRC inhibitor
in people. But so far it seems safe, he said.

Meanwhile, Novartis also designed its own new compound, AMN107,
dubbed "super Glivec," to overcome the weaknesses in Gleevec. It is
in Phase I safety tests in 76 patients with advanced leukemia and has
shown a strong response in half of them, said Dr. Francis Giles of
the M. D. Anderson Cancer Center in Houston.

"The response rates were dramatic," Giles said in a telephone
interview after presenting his findings to the hematology meeting.
Giles and colleagues started patients on very low doses of AMN107,
but he believes higher doses will show even better responses.

AMN107 is up to 30 times more potent than Gleevec because it was
designed to more efficiently bind to the BCR-ABL enzyme, Giles
said. "Clearly, we have a decent shot at a cure," he said.

Freeda:

I am getting Procrit shots once a month for my anemia.  I have had
lots of problems with fatigue.  It is okay to take Procrit for
anemia if you have CML and are on Gleevec.  Lots of people do.  I
know there was a study done on this but I can't remember where I
read about it.


My oncologist also suggested that I take Flintstones vitamins with
iron.

I still sometimes have to take a nap in the afternoon even with the
Procrit shots.  Also if I do too much I pay for it later.

WASHINGTON (Reuters) -- A next-generation leukemia pill designed to help
patients not cured by the successful drug Gleevec works even better than
doctors had hoped, researchers said Sunday.

The new drug, made by Bristol-Myers Squibb put 86 percent of patients who
tried it into remission -- meaning signs of their cancer disappeared, the
researchers said.

Although this was only a Phase I trial meant to show the drug was safe, the
effects were dramatic, the doctors told a meeting in San Diego of the
American Society of Hematology.

"Certainly it is wonderful. It will save lives," said Dr. Alan Kinniburgh,
senior vice president of research for The Leukemia and Lymphoma Society,
which helped sponsor the study.

Rumors of the new drug's success have been leaking for months because cancer
experts are so excited by the results. Oncologists hope the approach may
work in many other cancers, too.

The new drug is being tested in patients with chronic myeloid leukemia,
which affects about 4,400 Americans a year and 10,000 people around the
world.

The drug is known by its experimental name BMS-354825. During the trial,
also financed by Bristol-Myers, 31 of 36 patients with advanced CML who had
not been helped by Gleevec had a complete hematologic response, meaning
their bodies stopped producing leukemia cells.

This translates to an 86 percent remission rate, said Dr. Charles Sawyers, a
Howard Hughes Medical Institute investigator at the University of California
Los Angeles who is helping test the drug.

Gleevec, made by Swiss drug company Novartis, targets an enzyme called
BCR-ABL that leukemia cells use to proliferate. It attaches to the cancerous
cells and stops them from growing and spreading.

Targeted cancer pill
Sold in Europe under the name Glivec, it was the first "targeted" cancer
drug. It made headlines when it was approved in 2001 because never before
had a simple pill shown such dramatic effects in cancer.

Later Sunday Novartis will report on its new compound AMN107, dubbed "super
Glivec", also designed to overcome the weaknesses in Gleevec.

Gleevec, or imatinib, is Novartis's second-biggest product, with sales in
the first nine months of this year of $1.1 billion.

But in a few patients, perhaps 12 percent, the cancer cells mutate just
enough to slip out of Gleevec's grip. The cancer comes back.

So some of the researchers who worked on Gleevec teamed with Bristol-Myers
Squibb to develop the new pill, which is less picky about how it grabs onto
a cancer cell to deactivate it.

The Bristol-Myers drug affects a different enzyme called SRC, pronounced
"sark".

"It is definitely not Gleevec," Sawyers said in a telephone interview. "It's
not Gleevec 2. It has a completely different chemical structure."

Sawyers said he and colleagues worried that there could be unforeseen
side-effects in patients, as no one had ever tested a SRC inhibitor in
people. But so far it seems safe, he said.

There are more than 17 known mutations in leukemia cells that allow the
cancer to evade Gleevec's effects, but most of them seem to be susceptible
to BMS-354825, Sawyers said.

One mutation, known as T315I, resists both Gleevec and the Bristol drug.
"That mutation will likely require a different drug, and researchers are
working on that now," said Sawyers.

Kinniburgh said it is likely that the new pills will be used in combination,
like old-fashioned chemotherapy.

"I know of no cancer where one single drug has ever cured the cancer," he
said.
