I still have eight (8) bottles "on-loan" to Third Day Resources.
As soon as I go to my MD for my periodic check-up, I am going to try
some other alternative herbal therapies to augment the Combetic.
I'll let you know how that works out.
Between a brutally Spartan and strict diet, skimping on the Combetic,
and the other alternatives, I can keep my blood glucose at acceptable
levels. I wish there were ways which we could "grow our own" herbal
ingredients that are contained in Combetic to get away from having to
import everything from an unreliable trading "partner" like China.

I think you will find that we are all different. I took metformin
(Glucophage) for a very short period of time, and then absolutely
refused to take it any more, as it does do liver damage, regardless
of what they tell you. You have to weigh the risks for yourself. The
MD's have tried to get me onto other oral meds but I am
"uncooperative". I am now using only Combetic and and a twice daily
injection of slow-acting Lantus insulin (25 ml). I also take a number
of supplements, but not like I did before I started with Combetic. My
h1Ac readings over the past year since starting Combetic have been
from a low of 6.0 to as high as 7 (during a period of time I had a
long term sinus infection and pneumonia), and most recently back to
6.4, so both my doctor (I had to change doctors) and I are happy. I
also have fibromyalgia, which does affect my blood sugar levels
significantly.

Southeast Asian herb, has been reported to affect numerous activities

compound by ethanol extraction, distribution in n-butyl
alcohol/water, solid phase extraction/separation, and several rounds
of reverse phase high pressure liquid chromatography. We have shown
by NMR and mass spectrometry that this active compound is a novel
saponin, a gypenoside, which we have named phanoside (21-,23-epoxy-
,3beta-,20-,21-trihydroxydammar-24-ene-3-O-([alpha-d-rhamnopyranosyl
(1-->2)]-[beta-d-glycopyranosyl(1-->3)]-beta-d-lyxopyranoside)), with
a molecular mass of 914.5 Da. Phanoside is a dammarane-type saponin,
and four stereoisomers differing in configurations at positions 21
and 23 were identified, each of which were found to stimulate insulin
release from isolated rat pancreatic islets. We have also found that
the stereoisomers are interconvertible. Dose-dependent insulin-
releasing activities at 3.3 and 16.7 mM glucose levels were
determined for the racemic mixture containing all four stereoisomers.
Phanoside at 500 microM stimulates insulin release in vitro 10-fold
at 3.3 mM glucose and potentiates the release almost 4-fold at 16.7
mM glucose. At these glucose levels, 2 microm glibenclamide
stimulates insulin release only 2-fold. Interestingly, beta-cell
sensitivity to phanoside is higher at 16.7 mM than at 3.3 mM glucose,
although insulin responses were significantly increased by phanoside
below 125 microM only at high glucose levels. Also when given orally
to rats, phanoside (40 and 80 mg/ml) improved glucose tolerance and
enhanced plasma insulin levels at hyperglycemia.

Gynostemma pentaphyllum is a traditional Chinese medicine used for a
variety of conditions, including elevated cholesterol. We have
examined the pharmacological anti-hyperlipidemic and hypoglycemic
effectiveness of Gynostemma pentaphyllum in the obese Zucker fatty
diabetic rat model. After treatment for 4 days Gynostemma
pentaphyllum 250 mg/kg reduced triglyceride (33%), total cholesterol,
(13%) and low density lipoprotein cholesterol levels (33%). These
effects were dose-dependent and maintained for at least 5 weeks.
Chronic treatment for 3-5 weeks also reduced post-prandial
hypertriglyceridemia induced by olive oil 10 mg/kg in the Zucker
fatty rats but had no significant effect in lowering sucrose-induced
hyperglycemia in Sprague-Dawley rats. A novel regulation by
Gynostemma of glucose levels was also observed in the Zucker fatty
rat model. In a glucose tolerance test in obese and lean Zucker rats
pretreatment with Gynostemma pentaphyllum 250 mg/kg demonstrated
glucose levels were significantly less 2 hours post challenge (20%)
in the Gynostemma pentaphyllum obese rats compared to the control
group. Gynostemma pentaphyllum did not significantly reduce glucose
levels at 120 min in the lean strain, in contrast to the 20% decrease
seen in the obese rat. In vitro, Gynostemma pentaphyllum inhibited
alpha-glucosidase activity (50% inhibition at 42.8), which compared
to acarbose (50% at 53.9 microg/mL). The improvement in glucose
tolerance at 120 min by Gynostemma pentaphyllum in obese Zucker fatty
rats but not lean rats suggests that it may improve insulin receptor
sensitivity and together with the significant reduction of
hypertriglyceridemia, cholesterol and low density lipoprotein
cholesterol suggests that Gynostemma should be examined further by
oral hypoglycemic/anti-hyperlipidemic therapy.

The effect of water extract of Gynostemma pentaphyllum was evaluated
on 6-month chronic toxicity in Wistar rats. Control group received
orally 10 ml kg(-1) day(-1). The extract was orally given to the five
treatment groups at the doses of 6, 30, 150, 750 and 750 mg kg(-1) day
(-1) for 24 weeks. The last group served as the recovery group. The
results showed that the extract did not produce any significant dose-
related changes. Therefore, it is concluded that the extract of G.
pentaphyllum at the given doses did not produce any significant toxic
effect in rats during 6-month period of the treatment.
JIAOGULAN OR GINSENG?
According to Subhuti Dharmanda, Ph.D., Portland Oregon, January 1999:
As a result of my investigations, jiaogulan became part of the
clinical treatment programs at our organization's clinics,
particularly for the treatment of persons undergoing cancer
therapies. Jiaogulan is a tonic adaptogen herb that can help protect
the body from the stresses of the cancer therapies.
Jiaogulan has been a relatively obscure folk remedy in China, mainly
used for treating lung diseases. It was recently discovered to
contain the same kind of active constituents as ginseng. In fact, a
few of its ingredients were shown to be identical to those found in
ginseng.
Ginseng became famous because of the root's shape, which is sometimes
like a human figure. To be an effective health-care product, the root
has to be grown for many years (6 years is the standard for most
cultivated ginseng ), which contributes to the rarity and high cost
of ginseng. Jiaogulan grows wild over a large region, and provides
low cost supplies of the active constituent from its leaves that can
be harvested each year. For this reason, jiaogulan is a very
appealing ginseng substitute.
Various Chinese clinical trails have shown that jiaogulan is safe and
has both chemical constituents and herbal properties comparable to
that of ginseng. Proponents of the herb suggest that it might even be
superior to ginseng, having a broder range of the active constituents
( more variety of saponin glycosides, Jiaogulan having 82, ginseng
having 28 ), and producing less of a stimulatory action in persons
who are sensitive to ginseng in that way. The body of research
supporting the use of jiaogulan is extensive and expanding, so much
so that jiaogulan has become a permanent addition to the valuable
collection of Chinese herbal tonics used as adaptogens.

Michael Blumert and Dr. Jialiu Liu:
Jiaogulan has been used by the people in the mountainous regions of
Southern China as an energizing agent. They take it as a tea before
work to increase endurance and strength, and after work to relieve
fatigue. It is taken for general health and as a rejuvenating elixir.
The local Chinese called Jiaogulan xiancao the "Immortality Herb,"
and described it, "Like ginseng but better."
Jiaogulan contains saponins, flavones, polysaccharides, amino acids,

components. The type of saponins of both Jiaogulan and Panax ginseng

Out of the 82 saponins or gypenosides of jiaogulan, 4 are exactly the

An international team of researchers has finally managed to locate
stem cells in the pancreas -- in mice, at least and could pave the
way for dramatic new therapies for diabetes, namely the regeneration
of beta cells so the body could once again produce its own insulin.
If confirmed in humans, it could mean new therapies. Until now,
scientists had all but abandoned hopes that the pancreas made its own
stem cells because they had failed to find evidence to support the
theory.
"This is the first conclusive evidence that there are stem cells in
the pancreas, but any potential benefit is a very long way away,"
said Juan Dominguez-Bendala, director of Stem Cell Development for
Translational Research at the Diabetes Research Institute of the
University of Miami Miller School of Medicine.
"If this kind of cell and their progenitors with a capacity to divide
exist in the pancreas of man, and if we can identify the factors that
are responsible to induce their proliferation and differentiation,
then these latter processes might be stimulated in vitro but also, by
noninvasive means, in vivo," said senior study author Harry Heimberg,
an associate professor at the Diabetes Research Center of Vrije
Universiteit Brussel in Belgium.
Diabetes is primarily a failing of the pancreatic beta cells to
produce insulin. Beta cells are one of several types of cells making
up clusters of cells called the islets of Langerhans. Insulin is a
hormone that moves blood sugar from the bloodstream to the cells,
where it is used as energy.
Islet-cell transplantation, in which islets are transferred from one
person to another, is performed today but is limited in scope because
of a shortage of donors, according to the study.
The very existence of pancreatic stem cells is controversial. A
recent study out of Harvard found that the major source of new beta
cells in adult mice was preexisting beta cells, not stem cells. The
finding reduced the urgency to track down pancreatic stem cells.
"If stem cells didn't contribute, what was the point," said Dominguez-
Bendala.
For this study, Heimberg and his colleagues cut off the duct that
drains digestive enzymes from the pancreas in mice. Within two weeks,
the number of beta cells in the pancreas doubled. Not only did the
number of beta cells increase, the mice started producing more
insulin.
"When damaged a specific way, it triggered stem cells" production,
Dominguez-Bendala said.
The newly identified stem cells were almost identical to embryonic
beta cell progenitors. In fact, the gene Neurogenin 3 (Ngn3), which
plays a role in embryonic development of the pancreas, is also
involved in the formation of these new beta cells, the researchers
said.
"This is a model of regeneration no one has tested before," Dominguez-
Bendala said. "From a basic science point of view, it's very
exciting. It opens the door to potential therapies. If we could
trigger regeneration, that would be fantastic."
"This demonstrates a stem cell repair mechanism in the pancreas that,
if we understand it more, then we can help develop more cures with
either transplantation or with drugs that can increase the body's own
stem cells and beta cells," said Paul Sanberg, director of the
University of South Florida Center for Aging and Brain Repair in
Tampa.
The findings are published in the Jan. 25 issue of the journal Cell.

Israeli 'guided missiles' induce cancer cells to commit suicide By Nicky Blackburn November 21, 2007
Target-In CEO Dr. Shai Yarkoni: We take the small molecule, fuse to it a protein that causes cell suicide and get a molecule that preserves the specificity of the target while making the killing very effective.
For a moment, Dr. Shai Yarkoni, president and CEO of biotechnology start-up Target-In, appears to veer off on a tangent. "There's something very interesting in the terms we take from the military to the medical world and vice versa," he says contemplatively. "Surgical strike, magic bullets, these are all phrases that pass from one industry to the other."
People call what Target-In is doing 'guided missiles', and in a way that's exactly what the company's technology is: a guided missile that can recognize and kill inflamed cells from tumors and other diseases. But Target-In's technology is more sophisticated than that. The technology doesn't just shoot diseased cells down, it makes them commit suicide.
Target-in was founded in 2002 and stems from over 10 years of academic research by Prof. Hiya Galski, a leading academic in the field of immunotoxins at Hebrew University, and Yarkoni, who was her PhD student. The pair had been looking for a human toxin that could be used to kill off diseased human cells.
The problem was, however, that no creature on the planet will create a toxin for itself because basically it's suicide to do so. Instead doctors were using alternative man-made toxic compounds like radioactivity, chemotherapy and antibodies to treat diseased cells. Then the researchers had a bright idea.
Cell apotosis (programmed cell death) is one of the pivotal mechanisms through which an organism can control its cells. It enables an animal or human to induce suicide in cells that no longer serve appropriately and is a physiological, rather than pathological response. It is also used actively in the development of a fetus. Yarkoni holds out his arm as an example.
"As long as it the shaft of the arm that is developing, everything is fine. But what happens when you get to the fingers?"
He touches the creases of skin between his fingers. "In the fingers the cells have to keep on growing, but where the creases are it has to stop. The way it does this is through the cells committing suicide.
"The whole idea of modern medicine is to imitate the processes that happen naturally in the body. In one bright moment we realized that the cell suicide mechanism is the toxic compound we were looking for," Yarkoni told ISRAEL21c.
Targeted cell death is only possible because of advances that have taken place in targeted therapies over the last few years. Antibodies, hormones and growth factors, are already being used to treat diseases by targeting the specific cells that cause the illness. The only problem with these therapies, however, is that they don't kill the cells and have little biological effect.
Target-In aims to change that. The company has so far created eight molecules that can be combined with the molecules of proven targeted therapies to create a single small protein with two functional domains.
"This protein acts through using the targeting molecule to get to the right cells, and then once there, initiates the physiological effects of cell suicide," explains Yarkoni.
"Think of it like a key in a keyhole," he adds. "Antibodies match certain receptors of the cell the way a key matches a key hole. But getting a key into a hole doesn't always open the door. We have to add something to make it more effective and kill the cells. We take the small molecule or key, fuse to it a protein that causes cell suicide and get a molecule that preserves the specificity of the target while making the killing very effective. And it does this with little, if any, side effects and no effect on safety."
The apotosis protein will not destroy healthy cells, for example, because outside the cell it cannot have any impact. The only way for it to enter the cell is through the targeting molecule. In addition only small amounts of the protein are needed. "We imitate the physiological process and the cell doesn't know that the process is artificial," says Yarkoni. "There's no collateral damage."
This all sounds very innovative, but the company's goal is not to waste time pioneering anything that doesn't need to be pioneered. As a result, the company plans to only use antibody targets that have already been proven by other companies - treatments like Herceptin, an antibody used to treat breast carcinoma cells. This treatment sells for more than $2 billion, according to Yarkoni, but doesn't cure the patients.
"It merely prolongs the remission time," he said. "The market of targeted therapies has to be upgraded and we are bringing the solution."
The company has now taken five prototypes through successful animal studies which indicate that the treatment is a generic solution. The company's lead compound, the IL2-Caspase3, now has full proof of concept. In studies this receptor proved to have a clear beneficial effect on animal models of Crohn's disease and Diabetes Type 1. This receptor is currently used in three products on the market - two antibody treatments, Simulict and Zenapax, and one immunotoxin, Ontak, which are used to treat cancer and transplantation rejection.
"The existing treatments suffer major drawbacks," says Yarkoni. "The antibodies have low efficacy and the immunotoxin has high toxicity inducing many side effects."
The company now plans to add its protein to these three compounds and test them in a clinical setting. Trials will begin in one year. The goal, to show that the new drug is safer than Ontak, and more efficacious than the antibodies.
To do this as quickly and efficiently as possible, Target-In has signed two licensing agreements with an Indian active pharmaceutical ingredient (API) company, Century Pharmaceutical, which develops anti-allergens and asthma therapies; and a US company based in Palo Alto in San Francisco, which is developing a product for leukemia. Target-In will co-develop its treatment with Century Pharmaceuticals.
Yarkoni, who worked as CTO at Collguard before joining Target-In, is expecting to receive fast-track status from the FDA to develop the new treatments at a faster rate than normal. This will enable the company to carry out Phase I clinical trials and then pass directly to a pivotal study with fewer patient numbers than normal Phase II and III trials.
Once the first treatments have gone through clinical trials and proved safe, the company intends to expand the indications to address other diseases.
Target-In is a small company employing six; most of the work is sub-contracted out. It received $200,000 in seed money from the Hebrew University of Jerusalem. "This was very unusual," says Yarkoni. "It was the first time that the university had invested money directly into a company. The head of Yissum Technology Transfer company (the commercial arm of the Hebrew University), strongly believed in us."
Since then the company has raised more than $4 million from private investors, and is now gearing up for its second round. The aim is to raise $10 million. Already Century Pharmaceutical has pledged $1 million, and Yarkoni says the company has also received a firm commitment for a further $3 million from a Japanese company. Further negotiations are taking place with US and Japanese venture capital companies.
This money will get the company through two clinical trials and proof of concept.
"For me this is déjà vu," says Yarkoni. "I remember the antibodies industry in the late '80s. No one believed in it. In 1997 the whole worldwide market of antibodies was $250 million. In 2007, it's worth $20 billion. I think this is going to happen once again. We expect to revolutionize the whole concept of targeting therapy."

I just recieved 3 bottles of Pansulin from Lois for which I thank you. I am going to take 6 Pansulin in place of the 6 Combetics that I was taking. I am trying that for a week and let you know how it works for me. As you know WITHOUT Combetic my numbers are in the neighborhood of 200 plus or minus 10. WITH Combetic my numbers have always been in the neighborhood of 85 to 110 which I thought to be adequate control. Now that Combetic is no longer in adequate supply, I am starting the new regimen of 2 tabs in the morning - 2 tabs at noon and 2 tabs in the evening. I hope that this new Pansulin will provide similar control.
